Product Specific Information

  • Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest.

The structure of p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 functions as a key regulator preventing proliferation of damaged cells and acts as an anticancer agent. Mutations in the TP53 gene are common in human cancers and cause loss of tumor suppressor activity. Germline mutations are also linked to cancer-prone syndromes such as Li-Fraumeni syndrome.
