Product Specific Information
This CD25-4E3 monoclonal antibody reacts to human CD25, also known as the low-affinity interleukin (IL)-2 receptor alpha. CD25 is expressed on activated T cells (including regulatory T cells), B cells, and macrophages. It associates with CD122 (IL-2 receptor beta chain) and CD132 (common gamma chain) to form the high-affinity IL-2 receptor. IL-2 receptor signaling is linked to lymphocyte proliferation and survival. CD25 serves as a marker for Hodgkin’s disease and is associated with autoimmune diseases such as Type I diabetes.
This monoclonal antibody is cross-reactive to rhesus monkey.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.06 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test (empirically determined).
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD25 (IL2 receptor alpha chain/IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The IL2 receptor is a heterotrimeric complex composed of IL2RA, IL2RB, and IL2RG. Homodimeric IL2RA chains form low-affinity receptors, while IL2RB homodimers yield medium-affinity receptors. IL2 expression in mature thymocytes is monoallelic, serving as a unique regulatory mechanism. IL2, primarily produced by mature T cells, acts as a growth, differentiation, and cell death regulator. It promotes B cell proliferation, enhances NK cell activity, and inhibits granulocyte macrophage colony formation. Mutations in this gene are linked to interleukin 2 receptor alpha deficiency and autoimmune disorders.
