Product Specific Information

This peptide corresponds to 16 amino acids near the amino terminus of human IL-27.
PEP-0359 can be used as a blocking peptide with polyclonal antibody PA5-20240.

Target Information

IL-27, a member of the IL-12 family, is a heterodimeric protein consisting of the p40-related protein Epstein-Barr virus-induced gene 3 (EBI3) non-covalently linked to an IL-12p35-related protein, p28 (also known as IL-30).
IL-27 is produced by activated APCs and mature dendritic cells. It exerts its activities on NK cells and naive CD4+ T cells. Expression analysis of IL-27 receptor components (WSX-1/TCCR and gp130) suggests IL-27 may also target mast cells and monocytes.
Binding of IL-27 to WSX-1/gp130 activates JAK1, STAT1, and STAT3 phosphorylation. WSX-1/TCCR-deficient mice show impaired Th1 responses and higher susceptibility to L. monocytogenes infection, indicating Th1 responses require IL-27.
IL-27 alone cannot induce CD4+ T cells to differentiate into IFN-γ-producing cells, but it initiates Th1 responses by inducing T-bet and suppressing GATA-3. T-bet maintains IL-12Rβ2 expression after CD4+ activation.
Recent studies show IL-27 has antitumor activity, enhancing CTL granzyme B expression, augmenting CTL function, inhibiting tumor growth, and inducing regression of neuroblastoma tumors.
