Target Information

Matrix metalloproteinases (MMPs) are proteolytic enzymes that degrade connective tissue components. They share a conserved metal-binding active site and are inhibited by tissue inhibitors of metalloproteinases (TIMPs). MMPs and TIMPs regulate angiogenesis.

MMP-2 is synthesized as a 631 amino acid proenzyme activated by cleavage of the first 80 amino acids. It includes propeptide, catalytic, and hemopexin domains. MMPs collectively degrade extracellular matrix components such as collagens, fibronectin, laminin, and basement membrane glycoproteins. Structurally, MMPs possess a signal peptide, propeptide, and catalytic domain with a conserved zinc-binding site.

Functionally, MMP-2 is involved in tissue remodeling. Mutations in the MMP-2 gene are associated with Winchester syndrome and Nodulosis-Arthropathy-Osteolysis (NAO) syndrome. Two transcript variants encoding different isoforms have been identified.
