Target Information

Biological actions of insulin and IGF-1 are mediated by their respective cell surface receptor tyrosine kinases that regulate multiple signaling pathways through activation of phosphorylation cascades. Insulin/IGF-1 binding to the extracellular domain leads to autophosphorylation of downstream target proteins. These receptors differ in sequence in regions conferring specificity for their ligands and in intracellular signaling domains, resulting in distinct functional outcomes. Defects in the insulin receptor (IR) are linked to insulin resistance syndromes, while IGF-1R defects may cause growth retardation. Both signaling cascades play key roles in cancer development.
