Target Information

LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a tag for epigenetic transcriptional activation, acting as a corepressor. It contains a SWIRM domain, a FAD-binding motif, and an amine oxidase domain. LSD1 is part of histone deacetylase complexes and silences genes by demethylation. It oxidizes substrates via FAD to generate imine intermediates that are hydrolyzed. LSD1 demethylates mono- and tri-methylated Lys-4 of histone H3 and may repress neuronal genes. It requires RCOR1/CoREST for nucleosome demethylation and may also demethylate Lys-9 of histone H3, leading to derepression of androgen receptor target genes. Mutations in LSD1 can cause cleft palate, psychomotor retardation, and distinctive facial features.
