Product Specific Information
The AA4.1 monoclonal antibody reacts with mouse C1qRp, a 130–140 kDa C-type lectin-like type I transmembrane protein. The AA4.1 antigen is expressed on early B cell development stages in bone marrow, and detectable in adult spleen and fetal liver.
In combination with markers such as Thy-1, Sca-1, c-Kit, CD43, and CD24, AA4.1 expression helps segregate lymphoid-committed progenitors (CLP) into primitive and differentiated subsets. Co-expression with surface IgM and CD23 defines transitional B cell subpopulations:
- T1: AA4.1+/CD23-/sIgM^hi
- T2: AA4.1+/CD23+/sIgM^hi
- T3: AA4.1+/CD23+/sIgM^lo
AA4.1 is also expressed in cytoplasmic vesicles of endothelial cells, megakaryoblasts, and platelets. Clones AA4.1 and 493 recognize different epitopes of the same molecule.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse bone marrow cells. Use ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.
Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the 405 nm violet laser, emitting at 600 nm. Recommended filter: 610/20 bandpass. Use Super Bright Complete Staining Buffer (SB-4401) when using multiple Super Bright conjugates to reduce non-specific interactions.
Handling Notes:
- Light-sensitive; protect from light.
- Fixation: Use IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333) for up to 3 days at 4°C.
- Excitation: 405 nm; Emission: 600 nm; Laser: Violet

Target Information
The CD93 protein is a type I membrane glycoprotein originally identified as a myeloid cell-specific marker. Although once thought to be a receptor for C1q, it is now understood to mediate intercellular adhesion and apoptotic cell clearance. Its cytoplasmic tail interacts with moesin, linking it to the cytoskeleton and cytoskeletal remodeling.
