Product Specific Information

The CX5 monoclonal antibody reacts with mouse NKG2D, a lectin-like molecule expressed on both human and mouse NK cells. Mouse NKG2D binds to retinoic acid-inducible RAE-1 (α, β, γ, δ, ε) and minor histocompatibility molecule H60, and can costimulate NK activation receptors via DAP12/DAP10 adaptor molecules.
NKG2D is expressed by spleen and liver NK cells, NK1.1⁺ thymocytes, in vitro activated LAK cells, and a subset of splenic NKT cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes, ≤1.0 µg/test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to reduce non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

Natural killer (NK) cells are lymphocytes that mediate lysis of tumor and virus-infected cells without prior activation. They regulate humoral and cell-mediated immunity and express several C-type lectins involved in NK function.
The NKG2 gene family encodes type II transmembrane proteins with extracellular C-termini and C-type lectin domains. This family resides within the NK complex region containing multiple NK-specific lectin genes.
