Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule. CD11b, associated with integrin beta2 (CD18), is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. It serves as a receptor for CD54, CD102, CD50, and iC3b, playing a key role in cell-cell and cell-matrix interactions.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Recommended usage: 5 µL (0.6 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample prior to combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule forming a heterodimer with CD18 (integrin beta-2). Known as Mac-1 or CR3, it is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia.
CD11b/CD18 acts as a receptor for ICAM-1 (CD54), ICAM-2 (CD102), ICAM-4 (CD242), CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
Expression varies across tissues and cell types, increasing during monocyte maturation. CD11b is widely used as a microglial marker.
Dysfunction is associated with systemic lupus erythematosus and ITGAM-related susceptibility.
