Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule associated with integrin beta2 (CD18). It is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. CD11b functions as a receptor for ICAM family members (CD54, CD102, CD50) and iC3b, mediating crucial cell-cell and cell-matrix interactions.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.6 µg) per test (100 µL final volume).

Cell number: 10⁵–10⁸ cells/test (empirically determined).

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer Usage:

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures and minimized spillover/spread.

Target Information

CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) is a 165 kDa adhesion molecule forming a heterodimer with CD18 (integrin beta-2), known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia.
CD11b/CD18 serves as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating essential adhesion processes.
Expression varies across tissues and maturation stages, and dysfunction is linked to systemic lupus erythematosus (SLE) and ITGAM-related susceptibility.
