Product Specific Information

The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 acts as a ligand for CD80 (B7-1) and CD86 (B7-2), serving as a potent co-stimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and cytotoxicity of CD3-activated T cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells (5 µL per test; 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

The NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance.
Add 5 µL per sample to antibody cocktails or single-color controls (per 100 µL cell sample).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with minimized spillover and spread.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell surface glycoprotein of the immunoglobulin superfamily. It provides the critical co-stimulatory signal for T cell activation by binding CD80 and CD86 on antigen-presenting cells.
CD28 prevents T cells from entering anergic or apoptotic states. In mice, all CD4+ and CD8+ T cells express CD28; expression varies by thymocyte maturation stage.
Diseases associated with CD28 dysfunction include mycosis fungoides and Sezary’s Disease.
