Target Information
Ubiquitin exists either covalently attached to another protein or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond as a monomer (monoubiquitin), polymer linked via different Lys residues (polyubiquitin chains), or a linear polymer linked via the initiator Met (linear polyubiquitin chains).
Each linkage type has distinct biological roles:
- Lys-6-linked: DNA repair
- Lys-11-linked: ERAD and cell-cycle regulation
- Lys-29-linked: Lysosomal degradation
- Lys-33-linked: Kinase modification
- Lys-48-linked: Proteasomal degradation
- Lys-63-linked: Endocytosis, DNA-damage response, NF-κB signaling
- Linear (Met-linked): Cell signaling
Free (unanchored) polyubiquitin also functions in activation of protein kinases and signaling pathways.
