Target Information
Ubiquitin exists either covalently attached to another protein or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond as monoubiquitin or polyubiquitin chains linked through various lysine residues (Lys-6, Lys-11, Lys-29, Lys-33, Lys-48, Lys-63) or via the initiator methionine (linear polyubiquitin chains).
Each linkage type confers distinct cellular functions:
- Lys-6-linked: DNA repair
- Lys-11-linked: ERAD (endoplasmic reticulum-associated degradation), cell-cycle regulation
- Lys-29-linked: Lysosomal degradation
- Lys-33-linked: Kinase modification
- Lys-48-linked: Proteasomal degradation
- Lys-63-linked: Endocytosis, DNA-damage response, NF-κB activation
- Linear (Met-linked): Cell signaling
