Target Information
DOK6 (Docking Protein 6)은 p62 Dok 패밀리의 Dok-4/5 subclass에 속하는 새로운 세포 내 어댑터 단백질입니다.
이 단백질은 발달 중인 중추신경계에서 높게 발현되며, Ret와 함께 교감, 감각, 부교감 신경절 및 신장 발생 중 요관 싹(ureteric buds)에서 공발현됩니다.
Pull-down assay 결과, Dok-6의 phosphotyrosine binding (PTB) 도메인은 GDNF로 활성화된 Ret의 인산화된 Tyr(1062) 잔기와 결합함이 밝혀졌습니다.
또한, Ret의 리간드 활성화는 Dok-6의 C-말단에 위치한 티로신 잔기의 인산화를 유도하며, 이는 Src 의존적 기전을 통해 매개됩니다.
따라서 Dok-6은 Ret-Src 신호전달 경로의 기질로 작용합니다.
