Target Information
CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily.
Originally identified as a surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1, CD30 binds to its ligand CD30L (CD153) to mediate various biological effects such as proliferation, activation, differentiation, and apoptosis.
CD30 plays a critical role in:
- Pathophysiology of Hodgkin’s disease and other CD30+ lymphomas
- Thymic negative selection as a costimulatory molecule
- Expression in Hodgkin’s and Reed-Sternberg cells, non-Hodgkin’s lymphomas, virus-infected T/B cells, and activated normal T/B cells
Additionally:
- CD30 expression is found in T cells producing Th2-type cytokines
- Soluble CD30 (sCD30) serves as a marker of Th2 immune response
- TRAF2 and TRAF5 interact with CD30 to activate NF-κB signaling
- CD30 acts as a positive regulator of apoptosis, limiting autoreactive CD8 T cell proliferation and protecting against autoimmunity
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
CD30 is expressed by:
- Mononuclear cells in Hodgkin’s lymphoma
- Reed-Sternberg cells
- Most Anaplastic Large Cell Lymphomas (ALCL)
- Embryonal carcinomas
