Product Specific Information

This antibody detects endogenous levels of PAK1/PAK2 only when phosphorylated at Thr423.

Target Information

Three isoforms of serine/threonine kinases—designated αPAK (p68), βPAK (p65), and γPAK (p62)—exhibit high sequence homology with the S. cerevisiae kinase Ste20, which is involved in pheromone signaling.
The α, β, and γPAK isoforms specifically complex with Rac1 and Cdc42 in their active GTP-bound state, inhibiting intrinsic GTPase activity and leading to autophosphorylation.
There are eight autophosphorylation sites on γPAK, including Ser19, Ser141, and Thr402; phosphorylation of Ser141 and Thr402 correlates with γPAK activation.
Once phosphorylated, and with reduced affinity for Rac/Cdc42, the PAK isoforms dissociate from the complex to interact with downstream substrates.
