Beta Arrestin 1은 중추신경계에서 특히 풍부하게 발현되는 단백질 패밀리의 구성원으로, 어댑터 또는 스캐폴드 분자로서 작용합니다. 이 단백질은 mitogenic signaling에 필수적이며, G 단백질 연결 수용체(GPCR)의 작용제 의존적 탈감작 및 내재화를 매개합니다.
GPCR이 리간드와 결합하고 G 단백질과 상호작용하면, G-protein receptor kinases (GRKs)에 의해 세린 잔기에서 인산화됩니다. 세포질 내 Beta Arrestin 1은 ERK1/2에 의해 serine 412에서 인산화되어 음성 피드백 메커니즘으로 작용하며, 인산화된 수용체에 결합합니다. 이후 GPCR은 내재화되어 Ras, Raf, ERK1/2 신호 전달 경로를 활성화합니다.
