Product Specific Information

The RMST2-33 monoclonal antibody reacts with mouse ST2 (IL-33R, interleukin 1 receptor-like 1), a member of the IL-1 receptor family. ST2 forms a ligand-dependent dimer with IL-1R accessory protein, and IL-33/ST2 interaction induces TH2 cytokine production.
A soluble isoform of ST2 acts as a decoy receptor, blocking IL-33/ST2 signaling. ST2 is expressed by TH2 lymphocytes, mast cells, eosinophils, basophils, innate lymphocytes, smooth muscle, and endothelial cells, playing roles in host defense, allergy, and inflammation. IL-33/ST2 interaction is also reported to be atheroprotective.

RMST2-33 has been shown to bind and activate ST2 in vitro.
Applications reported include flow cytometric analysis. Tested on mouse bone marrow-derived mast cells at ≤0.5 µg/test.

Target Information

IL-33R (ST2) is a receptor for IL-33 and part of the IL-1 receptor family. Its extracellular domain interacts with IL-1RAcP and IL-33, activating downstream NF-kB and MAPK pathways via MyD88, IRAK1/4, and TRAF6. This signaling promotes cytokine release (IL-5, IL-6, IL-8, IL-13).
Two main isoforms exist:

• ST2L: Membrane-bound form
• ST2S: Soluble form (acts as a decoy receptor)

IL-33R is expressed on mast cells, basophils, eosinophils, ILC2, and Th2 cells. It is implicated in allergic responses, arthritis, atherosclerosis, and sepsis.
