Product Specific Information
This 9E12 monoclonal antibody reacts with human and mouse promyelocytic leukemia zinc finger (PLZF), a member of the BTB-POZ family of transcription factors. Expression of this transcriptional repressor in immune cells differs between mice and humans.
- In mice: PLZF is highly expressed in immature CD1d-restricted NKT2 and NKT1 cells, and a subset of γδ (Vγ1.1⁺Vδ6.3⁺) T cells. It is also found in non-invariant CD1d-restricted T cells and non-CD1d-restricted innate T cells.
- In humans: PLZF is expressed in NK cells, γδ T cells, CD4⁺ and CD8⁺ T cells, MR1-specific mucosal-associated invariant T cells, and MHC Class II-restricted T cells developing via thymocyte–thymocyte interaction.
- PLZF is expressed by innate lymphoid cells and progenitor cells. It exists as a homodimer or complex with PLZP and is involved in NKT cell development, NK cell function, cellular quiescence, growth suppression, and homeostasis maintenance.
- It also inhibits gene expression induced by retinoic acid receptor.
This 9E12 antibody stains mouse thymocytes brighter than Mags.21F7.
Tested Application:
Intracellular staining and flow cytometric analysis of mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Recommended use ≤ 0.125 µg/test in a final volume of 100 µL. Cell number can range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
This gene encodes a zinc finger transcription factor of the Krueppel C2H2-type family, containing nine Kruppel-type zinc finger domains at the carboxyl terminus.
The protein is nuclear-localized, involved in cell cycle progression, and interacts with histone deacetylase. Aberrant gene rearrangements at this locus are associated with acute promyelocytic leukemia (APL). Alternate transcriptional splice variants have been identified.
