Calsequestrin (CASQ) is a Ca²⁺-binding protein primarily located in the junctional sarcoplasmic reticulum of skeletal and cardiac muscle. The cardiac form, CASQ2, is encoded by a separate gene. CASQ2 functions mainly in buffering sarcoplasmic reticulum Ca²⁺ ions and also regulates the open probability of ryanodine receptor 2 (RyR2). Mutations in CASQ2 are associated with stress-induced polymorphic ventricular tachycardia (CPVT2), which can lead to cardiac arrest.
