Product Specific Information

The RPA-T8 monoclonal antibody recognizes the human CD8a molecule (32–34 kDa), expressed as CD8αβ heterodimers or CD8αα homodimers. CD8a is found on thymocytes, mature T cells, and NK cells, and binds MHC class I molecules, playing a role in T-cell development and activation.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells at 5 µL/test (0.5 µg/test).
  • Fluorochrome: Brilliant Violet™ 650 (BV650), emission at 649 nm, suitable for violet (405 nm) laser.
  • Buffer Recommendation: Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize polymer dye interactions.
  • Light Sensitivity: Protect from light to avoid photo-induced oxidation.
  • Fixation Compatibility: Compatible with IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54); not compatible with methanol-based fixation.
  • Excitation / Emission: 407 nm / 649 nm (Violet Laser)

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, essential for signal transduction, T-cell differentiation, and activation.
It functions as a co-receptor with TCR for MHC class I molecules, while CD4 interacts with MHC class II.
CD8 is expressed on T cells, NK cells, monocytes, and dendritic cells, existing as CD8αβ heterodimers or CD8αα homodimers.
Engagement with MHC-I/peptide complexes recruits Lck kinase, leading to cytokine production and cytotoxic T lymphocyte activation.
Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
