Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes through surface CD14.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated on normal human peripheral blood cells at 5 µL (1.0 µg)/test. A test corresponds to staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Violet™ 650 (BV650) emits at 649 nm and is optimized for cytometers equipped with a violet (405 nm) laser. Ensure compatibility before use.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to reduce non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light to avoid photo-induced oxidation.

Fixation Guidance:
Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark. BV650 is not compatible with methanol-based fixation.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and is released in soluble form (sCD14) in serum. CD14 participates in immune response modulation, apoptotic cell binding, and inflammatory processes. Dysfunction is associated with infections and respiratory conditions such as Croup.
