Reconstitution:
We recommend that the vial be briefly centrifuged prior to opening to bring the contents to the bottom.
Lyophilized human IL-17A should be reconstituted in sterile, distilled water to a concentration of 0.1–1.0 mg/mL.
For maximal stability, a carrier protein such as 0.1–1.0% BSA may be included.
The reconstituted protein should be apportioned into working aliquots and stored at ≤ -20°C.
Further dilutions should be made in low endotoxin medium or buffered solution with FBS or tissue culture grade BSA.
Storage:
For maximal stability, store this lyophilized preparation at ≤ -20°C, preferably desiccated.
For shipment and short-term storage (up to 2 weeks), storage at 2–8°C is sufficient.
Reconstituted human IL-17A should be stored at ≤ -20°C (not in a frost-free freezer).
Keep freeze-thaw cycles to a minimum.
Target Information
Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine that promotes inflammatory responses in cell lines and is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
IL-17A is a 32 kDa, disulfide-linked homodimer consisting of 136 amino acids, a member of a six-protein family (IL-17A–17F), signaling through the IL-17 receptor (IL-17R/CDw217).
High levels of IL-17A homodimer are produced by activated peripheral blood CD4+ T-cells.
IL-17A enhances expression of ICAM-1 in human fibroblasts and stimulates epithelial, endothelial, or fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2.
In the presence of IL-17A, fibroblasts can sustain proliferation of CD34+ hematopoietic progenitors and induce maturation into neutrophils.
Mouse, rat, and human IL-17A can induce IL-6 secretion in mouse stromal cells, indicating cross-species receptor recognition.
IL-17A regulates NF-kappa B and MAP kinase activities, stimulates IL-6 and COX-2 expression, and enhances nitric oxide (NO) production.
