This 15-414 monoclonal antibody reacts with human CD172a (signal regulatory protein alpha, SIRP alpha), a cell surface glycoprotein composed of three Ig-like extracellular domains and two cytoplasmic ITIMs. These motifs recruit SHP-1 and SHP-2 phosphatases. CD172a is expressed on monocytes, granulocytes, dendritic cells, and CD34+CD38- hematopoietic progenitor cells, but not on T or B lymphocytes. Neurons also express CD172a. Its ligand is CD47, and it is involved in dendritic cell-mediated T cell activation, neutrophil migration, and phagocytosis.

Target Information

SIRP alpha (CD172a) is a receptor-type transmembrane glycoprotein expressed on myeloid lineage cells (granulocytes, dendritic cells, macrophages, mast cells, hematopoietic stem cells). It serves as a substrate for activated tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and regulates receptor tyrosine kinase signaling negatively.

Binding with CD47 leads to ITIM phosphorylation and recruitment of SHP-1/2 phosphatases, mediating inhibition of macrophage phagocytosis and modulation of T cell/DC activation. SIRP alpha influences cellular responses to growth factors, insulin, oncogenes, and adhesion signals. High CD47 expression in cancer cells activates SIRP alpha, inhibiting macrophage-mediated tumor cell clearance.
