Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells.
SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting overlapping epitopes, while OKT3 and SK7 recognize different epitopes.
The antibody SK7 recognizes chimpanzee CD3.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits encoded on chromosome 11.
CD3 components have long cytoplasmic tails with double tyrosine-based motifs involved in signal transduction.
CD3 is present on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum. It is absent from B and NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
