Product Specific Information

The eBioG4.18 monoclonal antibody recognizes the rat CD3 protein, a critical component of the T-cell receptor (TCR) complex and a marker of T-cell lineage. CD3 mediates signal transduction upon TCR engagement, leading to cellular responses such as IL-2 transcription and clonal expansion.

In vitro, immobilized G4.18 activates T cells, while soluble G4.18 inhibits allogeneic mixed-lymphocyte proliferation and cytotoxicity. In vivo, G4.18 induces long-term specific tolerance to organ allografts.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of rat splenocytes at ≤0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal assay performance.

Target Information

The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating surface expression of the TCR complex. CD3 subunits (gamma, delta, epsilon, zeta) are part of the immunoglobulin superfamily encoded on human chromosome 11. These subunits contain cytoplasmic tails that associate with signal transduction molecules via tyrosine-based motifs.

CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Altered CD3 expression can occur in autoimmune diseases. Defects in CD3 genes are linked to CD3 immunodeficiency.
