Product Specific Information

This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios.
Helios is a zinc-finger transcription factor related to Ikaros, expressed mainly in hematopoietic stem cells and regulatory T cells. It is not expressed in mature B cells, dendritic cells, or myeloid cells.

Helios forms homo- and heterodimers with other Ikaros family members to regulate transcription. Eight isoforms exist, some with dominant-negative functions upregulated in lymphoid malignancies such as T-acute lymphoblastic leukemia.
22F6 may cross-react with rat Helios.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using mouse lymph node cells and Foxp3/Transcription Factor Buffer Set (Product #00-5523-00).

Use at 5 µL (0.015 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
APC-eFluor® 780 emits at 780 nm and is excited with the Red laser (633 nm). Ensure instrument compatibility.
Light-sensitive: protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific fixation performance should be determined empirically.

Target Information

Helios belongs to the Ikaros family of zinc-finger proteins, including Ikaros and Aiolos. These transcription factors regulate lymphocyte development and hematopoietic differentiation. Helios forms homo- or heterodimers with other family members. Multiple transcript variants exist; some may have unverified biological validity.
