Product Specific Information

The DB-1 antibody can neutralize the bioactivity of natural or recombinant IFN-gamma.
This antibody has been well characterized for ELISA, immunohistochemical staining, and neutralization (in vitro and in vivo).

IFN-gamma, also called Type II interferon, is a homodimeric glycoprotein containing approximately 21–24 kDa subunits.
The human IFN-gamma gene, located on chromosome 12, contains three introns; four exons code for a polypeptide of 166 amino acids, including a 20-amino-acid signal peptide.
Production of IFN-gamma is restricted to T cells and NK cells, unlike IFN-alpha and IFN-beta which can be synthesized by many cell types.

IFN-gamma exhibits antiviral and antiparasitic activities and inhibits proliferation of various normal and transformed cells.
It synergizes with TNF-alpha and TNF-beta to enhance inhibition of cell proliferation.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor and immune interferon primarily produced by T-lymphocytes and natural killer cells in response to antigens, mitogens, and cytokines.
It is a dimeric glycoprotein composed of two 146 amino acid subunits (~45 kDa as a homodimer).
On SDS-PAGE, IFN gamma appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.

The biological activity of IFN gamma is species-specific; human IFN gamma does not cross-react with mouse.
Functions include:

  • Antiviral and antiproliferative activity
  • Induction of MHC class I and II
  • Macrophage activation
  • Enhanced immunoglobulin secretion by B lymphocytes

Activation occurs through IFN gamma receptor I and II and the JAK-STAT pathway.
Human IFN gamma shares ~40% sequence homology with mouse IFN gamma.
Expression is upregulated by IL2, FGF basic, EGF, and downregulated by vitamin D3 or DMN.
Mutations in the IFN gamma gene are associated with aplastic anemia.
