Despite its predicted molecular weight, PD-1 often migrates at a higher molecular weight in SDS-PAGE. A suggested positive control is THP-1 cell lysate.

Cell-mediated immune responses are initiated by T lymphocytes stimulated by peptides bound to MHC molecules on antigen-presenting cells (APC).
Activated T cells express receptors such as PD-1 (PDCD-1) that mediate inhibitory signals from APCs. PD-1 binds two ligands, PDL-1 and PDL-2, and upon binding, inhibits immune activation in the absence of danger signals such as LPS or pathogen-associated molecules.
PD1-deficient mice exhibit hyperactivated immune systems and autoimmune diseases.
