Target Information
Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), referred to as A-to-I RNA editing. This process can influence gene expression and function in various ways, including:
- Altering mRNA translation by changing codons and amino acid sequences
- Modifying pre-mRNA splicing by altering splice site recognition
- Affecting RNA stability and genetic stability of viral genomes
- Influencing microRNA production, targeting, and protein-RNA interactions
ADAR can edit both viral and cellular RNAs, either at multiple sites (hyper-editing) or specific sites (site-specific editing). Known cellular RNA substrates include BLCAP, GRIA2, HTR2C, and GABRA3, with site-specific editing leading to amino acid substitutions that alter protein function.
Viral RNA substrates include HCV, VSV, MV, HDV, and HIV-1, where ADAR exhibits proviral or antiviral effects depending on the virus and mechanism. For example:
- Inhibits HCV replication via RNA editing
- Enhances MV, VSV, and HIV-1 replication through editing-independent mechanisms
- Stimulates HIV-1 infectivity via editing of viral RNAs
- Modulates HDV replication through A-to-I editing of amber/W codon
High levels of ADAR1 can inhibit HDV replication.
