Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), known as A-to-I RNA editing.
This process can alter gene expression and protein function through changes in:
- mRNA translation (codon modification)
- pre-mRNA splicing
- RNA stability and nuclease recognition
- RNA virus genome replication
- microRNA processing and protein-RNA interactions
ADAR1 edits both viral and cellular RNAs, acting in either proviral or antiviral roles depending on the virus type.
Notable substrates include BLCAP, GRIA2, HTR2C, and GABRA3, where site-specific editing leads to amino acid substitutions and functional modulation.
ADAR1 influences replication of viruses such as HCV, VSV, MV, HDV, and HIV-1, with editing-dependent and editing-independent effects.
