Product Specific Information
The 6.7 monoclonal antibody reacts with human and non-human primate (rhesus or cynomologus) CD18, also known as beta2 integrin. CD18 is a 90–95 kDa type I transmembrane protein that heterodimerizes with CD11a, CD11b, and CD11c. The CD18/CD11a complex (LFA-1) plays a critical role in cellular adhesion and inflammatory reactions.
CD18 is expressed broadly by human lymphocytes and monocytes, with lower expression in granulocytes. The antibody 6.7 has been reported to induce proliferation of resting T cells cultured with monocytes.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human blood cells.
Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Excitation: 488 nm; Emission: 520 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD18 integrin beta 2 subunit is a 90 kDa type I transmembrane protein expressed on all leukocytes. It forms heterodimers with CD11 molecules (CD11a–d) to constitute leukocyte (β2) integrins:
- αLβ2 (CD11a/CD18, LFA-1)
- αMβ2 (CD11b/CD18, Mac-1, CR3)
- αXβ2 (CD11c/CD18)
- αDβ2 (CD11d/CD18)
These integrins are essential for proper leukocyte migration and intercellular adhesion.
Deficiency of CD18 leads to leukocyte adhesion deficiency type I (LAD1) and related pathologies.
CD18 is also a target of Mannheimia haemolytica leukotoxin, mediating cytolysis.
Two transcript variants encoding CD18 protein have been identified.
