Target Information
The Lipin family of nuclear proteins includes Lipin-1, Lipin-2, and Lipin-3, all containing a nuclear signal sequence, a conserved NLIP domain, and a CLIP domain. LPIN1 (Lipin-1) is essential for normal adipose tissue development and metabolism. It activates PGC-1α–dependent pathways such as fatty acid oxidation and mitochondrial oxidative phosphorylation through PPARA expression. LPIN1 also suppresses lipogenic programs and reduces circulating lipid levels. Excess LPIN1 promotes fat accumulation and insulin sensitivity, while deficiency can lead to lipodystrophy, fatty liver, and insulin resistance.
