Target Information

RIPK2 is a serine/threonine kinase involved in cell activation, proliferation, differentiation, and apoptosis. Upon stimulation by bacterial peptidoglycans, NOD1 and NOD2 are activated, oligomerize, and recruit RIPK2 through CARD–CARD domains. RIPK2 contributes to the tyrosine phosphorylation of ARHGEF2 via Src kinase, leading to NF-κB activation by NOD2. Once recruited, RIPK2 autophosphorylates and undergoes Lys-63–linked polyubiquitination mediated by E3 ligases XIAP, BIRC2, and BIRC3. This modification facilitates the recruitment of MAP3K7/TAK1 to IKBKG/NEMO, inducing further polyubiquitination and activation of IKBKB/IKKB. Activated NF-κB then translocates to the nucleus to promote transcription of genes related to immune response, growth control, and apoptosis protection. RIPK2 also plays a role in T-cell receptor signaling by promoting BCL10 phosphorylation and subsequent NF-κB activation.
