Target Information
Phosphofructokinase (PFK) is a major regulatory enzyme involved in sugar metabolism and energy production in all cells. In humans, three loci control PFK expression: M (muscle), L (liver), and P (platelet) subunits, variably expressed across tissues.
PFK forms tetramers through random association of these subunits, resulting in five possible isozymes (M4, M3L, M2L2, ML3, L4). It is an allosteric enzyme activated by ADP, AMP, or fructose bisphosphate and inhibited by ATP or citrate.
PFK catalyzes the key regulatory step of glycolysis. Deficiency can cause episodic illness with dark urine after exercise. Elevated red cell PFK levels are noted in individuals with Down syndrome.
