Target Information
Phosphofructokinase (PFK)는 세포 내 당 대사를 조절하는 주요 효소로, 에너지 생산에 핵심적인 역할을 합니다. 인간에서는 M(근육), L(간), P(혈소판) 세 가지 구조적 유전자가 존재하며, 조직에 따라 다르게 발현됩니다.
PFK는 두 개의 유전자 산물의 무작위 조합으로 이루어진 사량체(tetramer) 형태로 존재하며, 근육과 간에서는 각각 M4, L4 동질사량체로 구성됩니다. 적혈구에는 M4, M3L, M2L2, ML3, L4의 다섯 가지 이소효소가 존재합니다.
PFK는 ADP, AMP, fructose bisphosphate에 의해 활성화되고 ATP, citrate에 의해 억제되는 알로스테릭 효소입니다. 해당 과정(glycolysis)의 핵심 조절 단계를 촉매하며, PFK 결핍은 경미한 증상부터 생명을 위협하는 에피소드성 질환까지 다양한 형태로 나타날 수 있습니다. 대표적인 증상으로는 격렬한 운동 후 주황색에서 짙은 갈색까지의 간헐적 어두운 소변이 있으며, 다운증후군 환자에서는 적혈구 PFK 수치가 상승하는 것으로 알려져 있습니다.
