Target Information
Phosphofructokinase (PFK) is a major regulatory enzyme that catalyzes sugar metabolism, playing a key role in energy production for normal cell function. In humans, three structural loci control PFK: M (muscle), L (liver), and P (platelet) subunits, expressed variably across tissues. PFK forms tetramers composed of these subunits, resulting in five possible isoforms (M4, M3L, M2L2, ML3, L4). It is an allosteric enzyme activated by ADP, AMP, or fructose bisphosphate and inhibited by ATP or citrate. PFK catalyzes the key controlling step of glycolysis. Deficiency can cause episodic illness with dark urine after strenuous exercise. Elevated red cell PFK levels are observed in individuals with Down syndrome.
