The human Fcγ receptor (FcγR)-binding inhibitor is used to inhibit non-specific FcγR-mediated binding of mouse monoclonal antibodies during flow cytometric analysis of human tissue.
Four classes of Fcγ receptors have been identified: FcγRI (CD64), FcγRII (CD32), and FcγRIII (CD16). These receptors are expressed at varying levels across multiple cell lineages, with high expression in myeloid, granulocyte, and B cells.
FcγR functions include initiation of endocytosis, phagocytosis, and antigen presentation upon binding to host immunoglobulin. Mouse monoclonal antibody binding to human FcγRs varies depending on antibody isotype and clone.

Applications Reported:
FG Purified Human Fc Receptor Binding Inhibitor has been reported for use in flow cytometric analysis.

Usage Instructions:
To inhibit non-specific binding, add the inhibitor to samples and incubate on ice for 20 minutes. Without washing, proceed to stain with the primary antibody.

Applications Tested:
FG Purified Human FcγR-Binding Inhibitor has been titrated and tested by inhibiting fluorochrome-conjugated isotype control binding to U937 cells.
Use at 20 µL per test (defined as the amount of antibody staining a cell sample in a final volume of 100 µL).
Cell count per test may range from 10⁵ to 10⁸ cells, determined empirically.

Storage and Handling:
Use in a sterile environment.

0.2 µm post-manufacturing filtered.

Purity:

90%, determined by SDS-PAGE.

Endotoxin Level:
<0.001 ng/µg antibody (LAL assay).

Aggregation:
