Product Specific Information
The FNLAP monoclonal antibody reacts with human latency associated peptide (LAP, pro-TGF beta 1, LAP/TGF beta 1).
TGF beta is synthesized as a precursor containing LAP at the N-terminus and mature TGF beta at the C-terminus. Cleavage between amino acids 278 and 279 forms LAP and TGF beta dimers, which associate to form the small latent TGF beta complex.
LAP is secreted and found in the extracellular matrix, and can also be expressed on platelets and activated regulatory T cells due to binding with GARP (LRRC32).
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometric analysis of stimulated normal human peripheral blood cells.
Use at 5 µL (0.5 µg) per test; a test corresponds to staining in 100 µL final volume.
Cell number may range from 10⁵ to 10⁸ cells/test.
Excitation: 488–561 nm

Target Information
TGF beta-1 is a cytokine found ubiquitously in tissues. It is stored in the extracellular matrix as a latent complex with its prodomain. Activation requires binding of αV integrin to an RGD sequence and mechanical force on the domain, which is anchored by latent TGF-beta binding proteins (LTBP).
Latent forms are complexes of TGF-beta, the N-terminal portion of the precursor (TGF-LAP), and a specific binding protein (LTBP).
