Target Information

Activator of S phase kinase (ASK, DBF4) is a regulatory subunit of the human Cdc7 kinase complex, crucial for the G1/S cell cycle transition in mammalian cells. Increased Cdc7 kinase activity at the G1/S boundary correlates with elevated ASK expression in late G1. ASK interacts with replication origins, suggesting that Cdc7 may trigger S phase by activating replication initiation complexes. Human ASK expression parallels Cdc7 kinase activity, both involved in initiating DNA replication via MCM2 targeting. ASK localization and chromatin binding increase at late G1 phase, potentially requiring additional signals. A functional link may exist between menin (a tumor suppressor mutated in MEN1) and ASK in regulating cell proliferation.
