Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha)는 수용체형 막관통 당단백질로, 과립구, 수지상세포(DCs), 대식세포, 비만세포 및 조혈 줄기세포 등 골수계 세포에서 발현된다.
SIRP alpha는 여러 활성화된 티로신 키나아제(EGFR, PDGFR, src, insulin receptor 등)의 기질로 작용하며, 수용체 티로신 키나아제 신호 경로의 음성 조절에 관여한다.
CD47과 결합 시 SIRP alpha의 세포질 영역 내 ITIMs(면역수용체 티로신 기반 억제 모티프)가 인산화되어 SHP-1, SHP-2 티로신 인산화효소를 활성화한다.
이 상호작용은 대식세포의 식세포 작용 억제, T세포 및 DC의 양방향 활성화 등 다양한 조절 과정에 관여한다.
암세포는 CD47을 과발현하여 SIRP alpha를 활성화하고 대식세포에 의한 파괴를 회피하는 것으로 알려져 있다.
