Product Specific Information

Description:
The eBioCB16 monoclonal antibody recognizes CD16 (Fc gamma RIII), a low-affinity receptor for IgG (50–80 kDa). CD16 includes CD16A (FcγRIIIA) expressed in macrophages and NK cells, and CD16B (FcγRIIIB) expressed in neutrophils. Two polymorphisms (NA-1, NA-2) exist, with NA-2 homozygotes showing lower phagocytic activity. CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, initiating ADCC, phagocytosis, degranulation, and proliferation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using human peripheral blood cells at 5 µL (0.25 µg)/test.
A test is defined as the antibody amount for a 100 µL sample (10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 600 is excited by the 405 nm violet laser and emits at 600 nm. Recommended detection filter: 610/20 bandpass.
Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to minimize polymer interactions.

Light Sensitivity: Protect from light.
Fixation: Samples can be fixed using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C with minimal signal loss.

Target Information

CD16 (encoded by FCGR3A) recognizes the Fc portion of IgG, mediating immune complex clearance, cytotoxicity, and viral infection enhancement.
FCGR3A and FCGR3B are homologous genes on chromosome 1.

• FCGR3A: Expressed on NK cells (transmembrane-anchored)
• FCGR3B: Expressed on neutrophils (PI-anchored)
  Mutations are associated with Immunodeficiency 20, Herpes Zoster, and autoimmune susceptibility (e.g., SLE, neonatal neutropenia).
  Alternative splicing produces multiple isoforms.
