Description:
The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (CD22α and CD22β, 130 and 140 kDa) produced by alternative splicing. It is highly expressed on mature B cells and B cell lymphomas.

The extracellular region contains seven Ig-like domains capable of binding sialic acid-containing ligands. The intracellular portion contains six tyrosine residues within ITIM and activation-like motifs, which are phosphorylated upon B-cell receptor engagement, contributing to both positive and negative regulation of BCR signaling.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry on normal human peripheral blood cells. Use 5 µL (0.125 µg) per 100 µL test volume. Suitable cell numbers range from 10⁵ to 10⁸ cells/test.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed on B-lymphocytes. It appears early in B-cell differentiation alongside CD19 and is mainly expressed in mature B cells. CD22 is strongly expressed in hairy cell leukemia and can aid in phenotyping mature leukemias.

It binds preferentially to α2,6-linked sialic acid, and its recognition site can be masked by cis interactions. Upon ligand-induced phosphorylation, CD22 regulates B-cell receptor signaling, acting both as an activator and inhibitor through interactions with Src family kinases and cytoplasmic phosphatases.

CD22 is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
