Target Information

MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating p53 stability. It is located on chromosome 12q. Akt-mediated phosphorylation of MDM2 at Ser166 and Ser186 enhances its interaction with p300, promoting MDM2-mediated ubiquitination and degradation of p53. Phosphorylation also inhibits binding to p19ARF, further increasing p53 degradation. Overexpression of MDM2 can inactivate p53 and reduce its tumor suppressor function. MDM2 also has E3 ubiquitin ligase activity and is involved in apoptosis and cell cycle regulation. It is overexpressed in various human cancers, including soft tissue carcinomas and breast cancer. Additionally, MDM2 interacts with proteins such as retinoblastoma 1 and ribosomal protein L5, and more than 40 alternatively spliced transcript variants have been identified in tumor and normal tissues.
