Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are low and play a key role in allergic responses by binding to mast cells and basophils via the Fc epsilon receptor I, leading to histamine release. IgE is produced by plasma B cells and basophils and exists in both soluble and membrane-bound forms, both recognized by IgE21.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells. Recommended usage is 5 µL (0.125 µg) per test (100 µL final volume). Cell counts can range from 10⁵–10⁸ cells/test.

Super Bright 436 is excited at 405 nm and emits at 436 nm. Recommended detection filter: 450/50 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and compensation.

Target Information

IgE is one of five immunoglobulin classes in mammals, consisting of two heavy epsilon chains and two light chains. It plays a central role in type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding to Fc receptors on mast cells and basophils. Crosslinking of IgE induces degranulation, promoting allergic responses. IgE also contributes to parasite immunity.
