Target Information
Tuberous sclerosis complex (TSC)는 TSC1 또는 TSC2 종양 억제 유전자의 돌연변이에 의해 발생하는 상염색체 우성 종양 증후군입니다.
이들 유전자의 산물은 단백질 복합체를 형성하여 mammalian Target of Rapamycin (TOR) 신호 전달을 간접적으로 감소시킵니다.
TOR는 세포 성장과 세포 주기를 조절하는 진화적으로 보존된 serine/threonine kinase로, 영양 수준과 성장 인자 신호를 통합합니다.
TSC1/TSC2 복합체는 Rheb GTPase 활성을 자극하여 TOR 활성을 억제하며, TSC1 및 TSC2 과발현 시 Rheb에 결합된 GTP/GDP 비율이 감소하고 세포 성장이 억제됩니다.
40 kDa 크기의 짧은 TSC1 isoform도 존재하지만 그 기능은 알려져 있지 않습니다.
