Target Information
Epithelial sodium channels (ENaC) are amiloride-sensitive members of the Degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels. These channels possess two short intracellular amino and carboxyl termini, two membrane-spanning segments, and a large extracellular loop with a conserved cysteine-rich region.
ENaC isoforms (alpha, beta, gamma) are expressed in kidney, lung, and colon, playing a crucial role in sodium and fluid balance. ENaC mediates aldosterone-dependent sodium reabsorption in the distal nephron, regulating blood pressure.
Gain-of-function mutations in beta or gamma ENaC may cause Liddell’s syndrome (hypertension), while loss-of-function mutations in alpha or beta ENaC can lead to pseudohypoaldosteronism (PHA-1).
