Product Specific Information

This clone recognizes the L1 or Calprotectin molecule, an intracytoplasmic antigen composed of a 12 kDa alpha chain and a 14 kDa beta chain. Although initially described as binding epitopes common to both chains (Flavell et al., 1987), later studies showed that it detects an epitope expressed exclusively on the beta chain (Goebeler et al., 1994). The antibody reacts with granulocytes, monocytes, and tissue macrophages. Variable results have been reported for brain macrophages and microglia. The recognized epitope is well conserved, making this antibody suitable for detecting myeloid cells in multiple species.

IHC Notes:
This product requires protein digestion pre-treatment (trypsin or pronase) for paraffin sections.
Suggested positive control: Human spleen.
Flow cytometry recommendation: Use 10 µL of working dilution for 1 × 10⁶ cells in 100 µL.

Target Information

The S100A8/A9 heterocomplex (also known as MRP-8/MRP-14 or Calprotectin) constitutes ~60% of the cytoplasmic protein fraction in circulating polymorphonuclear granulocytes and is also found in monocytes, macrophages, and ileal tissue eosinophils.
It exhibits antibacterial, antifungal, immunomodulatory, and antiproliferative properties, and acts as a potent chemotactic factor for neutrophils. Plasma concentrations increase in conditions with elevated neutrophil activity, such as inflammatory bowel disease. Calprotectin can also be detected in feces, saliva, plasma, and synovial fluid during infection and inflammation.
