EGF 및 TGF-α 등의 리간드가 결합하면 EGFR은 이합체를 형성하고, 세포질 도메인 내 티로신 잔기의 자가 인산화를 유도합니다. 이러한 메커니즘은 두경부암, 뇌종양, 방광암, 위암, 유방암, 폐암, 자궁내막암, 자궁경부암, 난소암 등 다양한 암종에서 중요한 역할을 합니다.
