Product Specific Information

PA1-1055 detects phospho-IRS-1 (Ser636) in mouse and human cells.
This antibody has been successfully used in Western blot procedures.
By Western blot, it detects a ~170 kDa phospho-IRS-1 (Ser636) protein from 3T3-L1 cells treated with insulin for 10 minutes.

The immunogen is a synthetic phosphopeptide corresponding to residues D(631) Y M P M (pS) P K S V S A P Q Q I(646) of human IRS-1.
This peptide sequence is 100% conserved in rats and mice.
The peptide (Cat. # PEP-185) is available for neutralization and control experiments.


Target Information

IRS-1 is a major substrate of the insulin receptor and is phosphorylated in response to stimulation by insulin, IGF-1, and IL-4.
Phosphorylation occurs on serine, threonine, and tyrosine residues in various tissues.
Casein kinase II mediates part of the insulin-stimulated serine/threonine phosphorylation of IRS-1 in vivo.
Thr502 and Ser99 are identified as major phosphorylation sites catalyzed by casein kinase II.
IRS-1 contains three putative 14-3-3 binding sites (Ser270, Ser374, Ser641), with the motif around Ser270 located in the phosphotyrosine-binding domain responsible for insulin receptor interaction.
